Development of DGT passive sampler for the assessment of polycyclic aromatic hydrocarbons (PAHs) in water using AhR-CALUX as detection method

Polycyclic Aromatic Hydrocarbons (PAHs) are ubiquitous environmental pollutants[1, 2] generated primarily during incomplete combustions of organic materials (e.g. coal, oil, petrol, and wood). Anthropogenic emissions predominate in the environment and include residential heating, coal gasification and liquefying plants, carbon black as well as motor vehicle exhaust. PAHs are present in ambient air in the gas phase and in the form of aerosol, and eventually reach aquatic systems where they adsorb to particles. Many PAHs, as ligands to the aryl hydrocarbon receptor (AhR) [3, 4], have toxic or carcinogenic properties. Considering the low concentration of PAHs (ng·L-1) in aquatic systems [5-8], the major goal of this study is to develop and validate a novel long-term monitoring tool for measuring PAHs in aquatic systems, specifically in urban and industrial wastewaters. The method development involves the following steps: (1) the AhR-mediated activity evaluation of individual PAHs by the Chemically Activated LUciferase gene eXpression (CALUX) bioassay, (2) the DGT performance test using benzo[a]pyrene (BaP) as the model PAH, XAD 18 resin as the binding phase, (3) measurements of effective diffusion coefficient of BaP in agarose diffusive gel and comparison with theoretical calculating values, (4) the effects of pH, ionic strength and dissolved organic matter (DOM) on DGT trapping of BaP. Ultimately, this passive sampling combined with the use of AhR-CALUX bioassay will be applied in water and the results will be compared with conventional techniques.
