Description

The cell-free system for de novo synthesis for picornavirus (poliovirus, rhinovirus, TME virus) consists of an extract of uninfected cells and is programmed with viral RNA. Only viral molecules are synthesized, as the host RNA's are inactivated by a nuclease treatment. In this system upt o 10 to the 9th virions per ml are synthesized. Using this cell-free system, the goal of this project is to unravel the last steps of the replication cycle of poliovirus and to search for the targest of antiviral drugs. The cell-free system is an ideal tool for such studies, as there is direct access to the replication machinery, unhapered by intact cellular membrane(s).
Another goal of this project is to rebuilt the cell-free system for the synthesis of rhinovirus (common cold) and TME virus (another picornavirus, inducing a MS-like disease in mice).
