Cells of the immune system play a critical role in cancer and a range of inflammatory, infectious and neurological diseases and are the basis for innovative cell-based therapies. Purifying selected populations of immune cells from blood or the local tissue environment is crucial for their functional characterization and for the evaluation of cell-based therapies. Most cell sorters are based on a stream of fluid that is split into droplets under high pressure, which are then separated based on electric charges. This however puts a lot of stress onto the delicate immune cells and limits the amount and types of immune cells that can be viably isolated. To obtain high viability and optimal functionality of the sorted immune cells, we are hereby requesting funding for a MACSQuant® Tyto Cell Sorter, which uses a game-changing microchip sorting technology at low working pressure. Samples are kept contamination-free within its disposable, fully closed cartridges, which prevents aerosol and droplet formation, thus not only ensuring sterile storting but also providing a safe environment for the operator. The stand-alone benchtop system allows sorting of cells based on up to 10 different parameters, is easy to use and fully automated once the cartridge has been inserted. The system is not only very well suited for research, but it can also be used in a translational or clinical setting.
