Hematopoietic stem cells: characterisation of the subsets and enrichment for autologous transplantation .

Hematopoietic stem cells can be identified by their reactivity with the CD34 monoclonal antibody. Several subsets have been described according to their antigen phenotype and their behaviour in culture systems. In this project we will characterize these subsets in normal blood and bone marrow and in a variety of diseases with morphological, cytochemical, immunocytochemical and molecular biological techniques. The differentiation and maturation of these cells induced by cytokines in culture systems will be studied. Finally the subsets will be examined during mobilisation by chemotherapy and growth factors in patients with lymphoma and myeloma and in enriched stem cell suspensions prepared for autologous transplantation.
