The aims of the project are to investigate the use of immotile spermatozoa in ICSI, and to select, characterize and cryopreserve testicular spermatozoa for ICSI. So far, ICSI with immotile spermatozoa is less efficient than with motile spermatozoa, since motility is treated as a marker for vitality. Other parameters which might predict successful use of immotile spermatozoa will be studied. Vital immotile spermatozoa may be distinguished from non vital spermatozoa by various procedures, such as computer-assisted semen analysis or different hypo-osmotic swelling tests, which examine the integritiy of the sperm membrane. Ultrastructural analysis will be carried out to investigate the suitability of immotile sperm. To isolate spermatozoa from a testicular biopsy, several mechanical and enzymatical procedures will be compared. After Iysing the erythrocytes, the cell suspensions will be purified on discontinuous Percoll gradients, in order to obtain a pure spermatozoa fraction. The freezability of isolated testicular spermatozoa will be investigated, and preclinical ICSI experiments on research oocytes with the frozen-thawed spermatozoa will be carried out.
