Samenvatting

Objective
To understand the molecular mechanisms underlying the favourable effects of LH activity in ART on oocyte and embryo quality. Micro array analysis in mouse and human cumulus-corona cells (CC) indicated that Col4a1/2 and Col6a2 are upregulated in preovulatory follicles when stimulated with FSH alone compared to FSH plus LH-activity. Col4 expression is related to progesterone production and Col6 interacts with hyaluronic acid, an important extracellular matrix (ECM) constituent.

Design
Early preantral mouse follicles were individually grown in 10IU/L recombinant FSH (GONAL-F, Serono) for 12 days, or in a superovulation condition where follicles received from day 6 to day 12 (antral phase) either 100IU/L recombinant FSH or 100IU/L highly purified menotrophin (MENOPUR, Ferring Pharmaceuticals). CC cells were assayed on day 9 (early antral follicle) and day 12 (preovulatory follicle) for the amounts of Col4a1/2 and 6a2 mRNA.
