Samenvatting

Large population of thermophilic bacteria are naturally endowed with unique metabolic features, one of them is the PHB synthesis pathways. Thermophiles are more promising for PHB
production compared to mesophiles. As thermophiles metabolism operates at 50-90°C, avoiding the sterilization costs required for preventing contamination with mesophilic bacteria. In
addition, the high-temperature production limits cooling costs thereby enhancing economic feasibility.
Caldimonas thermodepolymerans, a moderate thermophile with the ability to form PHB granules between 45 °C to 55 °C, is studied for its potential use as a largescale producer. The bacterium
is engineered using fundamental physiological studies and molecular techniques. Regulator proteins are studied to establish a link between them and their expression levels and PHB
production, including those involved in manipulation of metabolic pathways, stress response, and the polymer production
