Samenvatting
Messenger RNA (mRNA) based therapeutics have been in the last 10 years extensively exploited as attractive treatment against cancer. However, in vivo delivery of in vitro transcribed (IVT) mRNA can cause rapid production of type I interferon and trigger a counteracting effect. On one hand a strong innate immune response in induced, on the other this is know to reduce mRNA translation and thus possibly decrease the mRNA vaccine efficiency. Here, we enhanced mRNA biological activity improving mRNA translatability in the cell and lowering the in-vivo molecular immunogenicity.
